METTL1介导的 m7G RNA 甲基化在肿瘤发生发展中的调控作用

The Regulation Function of METTL1 Mediated Methylation of m7G RNA in Tumor Development

摘要: RNA甲基化在许多基本的生物学过程中起着关键作用,包括基因表达的调控、信使RNA的稳定性和稳态。m7G甲基化通过修饰各种RNA分子,包括信使RNA(mRNA)、核糖体RNA(rRNA)、microRNA和转运RNA(tRNA)等,参与生理和病理功能。m7G是发生在tRNA可变环中最普遍的修饰之一,由人类METTL1-WDR4复合物调控。m7G修饰调控mRNA的转录、microRNA的生物合成、生物学功能、tRNA的稳定性、18SrRNA的加工和成熟。作为m7G的甲基转移酶,METTL1在各种肿瘤中发挥着重要作用,包括影响甲基化水平、肿瘤相关基因稳定性、癌细胞信号通路等多种调节机制。因此,阐明METTL1在肿瘤发生发展过程中的机理,对肿瘤的诊疗具有重要意义。本研究综述了METTL1介导的m7G RNA甲基化在急性髓细胞白血病(AML)、肝内胆管癌(ICC)、肝细胞癌(HCC)、结肠癌(CC)、肺癌、鼻咽癌、膀胱癌等各种肿瘤发生发展过程中调控作用的研究进展,探讨了METTL1与RNA稳定性的关系以及对多种肿瘤的调节机制,认为METTL1可能成为新的诊断标志物和治疗靶点。

Abstract: RNA methylation plays a key role in numerous fundamental biological processes, including the regulation of gene expression, the stability and homeostasis of messenger RNA. m7G methylation modifies various RNA molecules, such as messenger RNA (mRNA), ribosomal RNA (rRNA), microRNA, and transfer RNA (tRNA), which are involved in physiological and pathological functions. The tRNA variable loop is prevalently modified by m7G under the regulation of the human METTL1-WDR4 complex. This modification plays a crucial role in regulating mRNA transcription, microRNA biosynthesis, biological function, tRNA stability, 18SrRNA processing and maturation.As an m7G methyltransferase, METTL1 plays a crucial role in various tumors by modulating methylation levels, stabilizing tumor-related genes, regulating cancer cell signaling pathways and other regulatory mechanisms. Elucidating the mechanism of METTL1 in tumor initiation and progression is crucial for accurate cancer diagnosis and effective treatment. This study provides an overview of the research progress on the role of in METTL1-mediated m7G RNA methylation in various malignancies, including acute myeloid leukemia(AML), intrahepatic cholangiocarcinoma(ICC), hepatocellular carcinoma(HCC), colon cancer(CC), lung cancer, nasopharyngeal cancer, bladder cancer and other tumors. The discussion focuses on the relationship between METTL1 and RNA stability as well as the regulatory mechanisms of METTL1 in various tumors. These findings suggest that METTL1 could be developed as a new diagnostic marker and therapeutic target for tumors.
