单细胞测序技术的文献计量学研究及热点应用

摘要: 单细胞测序(single cell sequencing,SCS)作为新一代的测序方法,弥补传统bulk测序分析得出的基因平均表达量的不足,可在单个细胞水平上对特定的细胞亚群携带的遗传信息进行高通量的测序分析。一方面其在分子层面获得特定细胞亚群的基因序列、转录本、蛋白质及表观遗传学等信息,分析细胞亚群之间的遗传和蛋白质信息的差异,另一方面可获取不能直接培养的微生物等的遗传信息,可更好地理解它们在微环境中的特定作用等。单细胞测序技术不断发展,使得我们能更加准确地认识疾病的本质,对疾病的诊断、药物治疗效果监测以及预后的影响意义重大。本研究通过文献计量学的方法,对当前单细胞测序技术文献发表数目的趋势以及国家地区和关键字进行循证可视化,同时主要总结了单细胞测序的应用领域,包括在胚胎和器官发育、免疫系统、癌症进展、寄生虫和传染病、干细胞研究以及抗体筛选等方面的应用。

Abstract: Single cell sequencing (SCS), as a new generation of sequencing method, can make up for the shortage of the average gene expression obtained by the traditional bulk sequencing, and also can sequence the genetic information of the specific cell subgroups at single cell level. In terms of the advantages of SCS, it obtains the gene sequences, transcripts, proteins and epigenetics information of the specific cell subgroups and integrates and analyzes the differences between different clusters and subgroups. Meanwhile, it also obtains and understands the genetic information of the microorganisms that cannot be cultivated directly, to better understand their specific role in the microenvironment. With the advancement of SCS technology, it obviously allows us to understand the nature of diseases more accurately, which is of great significance to the diagnosis and prognosis of the disease, and assessment of the effect of drug treatment. This study visualizes the current trends in the number, countries, regions and keywords of SCS research publications through the bibliometrics analysis, and summarizes the application of SCS, which includes the application in embryos and organs, immune system, cancer progression, parasities and infectious diseases, stem cell research and antibody screening.
