电感耦合等离子体质谱法测定中药材中 15种金属元素的含量

Determining the Contents of 15 Metallic Elements in Traditional Chinese Medicinal Materials by ICP-MS

摘要: 目的:建立电感耦合等离子体质谱法(ICP-MS)测定中药材中锂(Li)、铍(Be)、钴(Co)、镍(Ni)、砷(As)、硒(Se)、锶(Sr)、锰(Mn)、镉(Cd)、锌(Zn)、铝(Al)、铊(Tl)、铅(Pb)、铋(Bi)、铜(Cu)15种金属元素的含量。 方法:取中药材样品,60 ℃烘干3 h,粉碎成粗粉。精密称取0.2 g,置消解罐内进行消解,最后用水定容至25 mL。建立ICP-MS方法测定样品中15种金属元素的含量,外标法定量。ICP-MS射频功率为1 150 W,雾化器氩气流速为1.20 L·min-1,等离子体氩气流速为15 L·min-1,采样深度为8 mm,反应模式为标准模式和碰撞模式。结果:在最佳仪器条件下,15种金属元素在0~100 μg·L-1范围内线性关系良好,相关系数均大于0.999,平均回收率为85.3%~114.7%,精密度的相对标准偏差(RSD)为0.68%~5.65%,检出限均低于0.047 μg·L-1,符合分析技术要求。结论:该方法快速、简便、重复性好、准确度高,可用于中药材中15种金属元素含量的测定。

Abstract: Objective:To establish a inductively coupled plasma mass spectrometry(ICP-MS)method for simultaneous determination of 15 metallic elements, Lithium(Li), Beryllium(Be), Cobalt(Co), Nickel(Ni), Arsenicum(As), Selenium(Se), Strontium(Sr), Manganese(Mn), Cadmium(Cd), Zinc(Zn), Aluminum(Al), Thallium(Tl), Plumbum(Pb), Bismuth(Bi), Cuprum(Cu),in traditional Chinese medicinal materials. Methods: Samples of traditional Chinese medicinal materials were dried at 60℃ for 3 h and crushed for coarse powder. Accurately weighed 0.2 g coarse powder of the sample,and placed into the digestion tank for digestion.Finally, the metered volume should be fixed to 25 mL by water. The ICP-MS method was established to determine the contents of 15 metallic elements, and external standard method was used for quantification. The instrument parameters of ICP-MS were as follows:radio frequency(RF) power at 1150 W, atomizer flow rate of argon gas at 1.20 L·min-1, plasma flow rate of argon gas at 15 L·min-1,sampling depth at 8 mm. The reaction modes were standard mode and collision mode. Results:Under the optimal instrument conditions, the linearity was good for 15 metallic elements in the concentration range of 0-100 μg·L-1, and the correlative coefficients was higher than 0.999.The recovery rate ranged from 85.3% to 114.7%. The RSD for precision ranged from 0.68% to 5.65%. The detection limits were all lower than 0.047 μg·L-1. The verified data meet the requirements of the analytical method. Conclusion: The method is rapid, simple, reproducible and accurate, and can be used for the content determination of 15 metallic elements in traditional Chinese medicinal materials.
