基于网络药理学探究复方丹参滴丸治疗冠状动脉粥样硬化性心脏病的作用机制

摘要: 目的:通过网络药理学方法探讨复方丹参滴丸治疗冠状动脉粥样硬化性心脏病(coronary artery disease, CAD)的潜在作用机制。方法:利用TCMSP和Swiss TargetPrediction数据库进行复方丹参滴丸化学成分潜在靶基因预测;通过GeneCards、OMIM和DisGeNET数据库检索CAD的相关靶点;运用Cytoscape软件进行网络可视化分析;Metascape平台对核心靶点进行GO以及KEGG通路富集分析。结果:复方丹参滴丸中丹参酮IIA、木犀草素、槲皮素、积雪草酸等56 种有效活性成分,可能通过作用于TNF、IL6、AKT1、VEGFA、IL1B等66个核心靶点,调控脂蛋白及动脉粥样硬化、HIF-1信号通路、TNF信号通路、PI3K-Akt信号通路等多种途径,起到抗炎、调节氧化应激及脂代谢紊乱、修复血管损伤等功效,从而达到治疗CAD的目的。结论:通过网络药理学方法预测了复方丹参滴丸治疗CAD的潜在靶点及作用机制,为后续基础研究提供了理论依据。

Abstract: Objective:To explore the potential mechanism of Compound Danshen Dripping Pills in the treatment of coronary arterydisease (CAD) based on network pharmacology. Methods:TCMSP and Swiss Target Prediction databases were used to predict the potential target genes of chemical constituents of Compound Danshen Dripping Pills. CAD relevant targets were retrieved through GeneCards, OMIM and DisGeNET databases. Cytoscape software was used for network visualization analysis. Metascape platform was used for GO and KEGG pathway enrichment analysis of core targets. Results:A total of 56 active components including tanshinone IIA, luteolin, quercetin, and asiatic acid and 66 key targets such as TNF, IL6, AKT1, VEGFA, IL1B were obtained. GO and KEGG analysis showed that Compound Danshen Dripping Pills may play the role of anti-inflammatory, regulate oxidative stress and lipid metabolism disorders, repair vascular damage through regulating lipoprotein and atherosclerosis, HIF-1 signaling pathway, TNF signaling pathway, PI3K-Akt signaling pathway and so on, to achieve the purpose of treating CAD. Conclusion:The potential target and mechanism of Compound Danshen Dripping Pills in the treatment of CAD were studied based on network pharmacology, which provided theoretical basis for subsequent basic research.
