基于文献分析的黄芩抗脑缺血损伤机制研究进展

    基于河间学派火热论的黄连解毒汤及其有效组分调控“中枢-外周”免疫互作治疗急性缺血性中风的多组学联动研究

摘要:

目的:通过文献挖掘分析黄芩抗脑缺血损伤的疗效机制,为临床运用黄芩治疗缺血性脑卒中(IS)后脑损伤提供依据。方法:检索20136月至20246月中国生物医学文献数据库(CBM)、中国期刊全文数据库(CNKI)、万方数据知识服务平台、维普网(VIP)和PubMed中收录的黄芩干预IS的相关文献,总结分析所涉及的作用机制及相关生物标记物。结果:共检索到64篇相关文献,符合本研究纳入标准的有10篇,其中9篇为动物实验研究,1篇为细胞实验研究,均为基础研究类文献。通过分析文献显示,黄芩抗IS后脑损伤的作用机制涉及抗炎、抑制细胞凋亡、抗氧化、减轻脑水肿、保护血脑屏障、促进神经元再生和抑制星形胶质细胞肿胀;相关的生物标记物主要有肿瘤坏死因子-αTNF-α)、超氧化物歧化酶(SOD)、水通道蛋白-4AQP-4)、丙二醛(MDA)、神经胶质纤维酸性蛋白(GFAP)、白细胞介素-IL-)、基质细胞衍生因子-1SDF-1)、谷胱甘肽(GSH)、G蛋白偶联受体30GPR30)、水通道蛋白-1AQP-1)、环磷酸腺苷(cAMP)、c-Jun氨基末端激酶(JNK)、p38丝裂原活化蛋白激酶(p38MAPK)、环氧化酶-2COX-2)、谷胱甘肽过氧化物酶(GSH-Px)、谷氨酸(Glu)、瞬时受体电位香草素受体4型通道(TRPV4)、Toll样受体4TLR4)、NOD样受体蛋白3NALP3)、细胞外调节蛋白激酶1/2ERK1/2)、5-溴脱氧尿嘧啶核苷(BrdU)阳性细胞。结论:黄芩抗IS后脑损伤疗效机制广泛,涉及抗炎、抑制细胞凋亡、抗氧化、减轻脑水肿、保护血脑屏障和促进神经元再生等,且具有途径广、靶点多、毒副作用小的特点,改善脑损伤效果显著。

Abstract:

Objective: To analyze the therapeutic mechanism of Scutellaria baicalensis on ischemic stroke IS brain injury through literature mining so as to provide the basis for clinical application of Scutellaria baicalensis in the treatment of brain injury after ischemic stroke.Methods: Literature from June 2013 to June 2024 related to Scutellaria baicalensis agnist ischemic stroke were retrieved from China Biomedical Literature Database CBM), China National Knowledge Infrastructure CNKI), Wanfang Data Knowledge Service Platform VIP and PubMed and the involved mechanism of action and related biomarkers were summarized and analyzed.Results: A total of 64 relevant studies were retrieved out 10 of which met the inclusion criteria including 9 animal experiment studies and 1 cell experiment study all of which were in the basic research category. Analysis of the literature showed that the mechanism of Scutellaria baicalensis against ischemic stroke brain injury involves anti-inflammation inhibition of apoptosis antioxidation alleviation of cerebral edema protection of the blood-brain barrier promotion of neuronal regeneration and inhibition of astrocyte swelling. The main related biomarkers are TNF-α SOD AQP-4 MDA GFAP IL- SDF-1 GSH GPR30 AQP-1 cAMP JNK p38MAPK COX-2 GSH-Px Glu TRPV4 TLR4 NALP3 ERK1/2 and BrdU-positive cells.Conclusion: The therapeutic mechanism of Scutellaria baicalensis on schemic stroke brain injury is extensive involving anti-inflammation inhibition of apoptosis antioxidation alleviation of cerebral edema protection of the blood-brain barrier and promotion of neuronal regeneration etc. and has the characteristics of wide range of pathways multi-targets and low toxicity and side effects which are effective in improving brain injury.
